I'll try to identify the important landmarks and the rest will be filled with my imagination.

Say I focus on a specific landmark (and surroundings) would the Dome of the Rock be a good pick?

While it is certainly iconic it's only important to Islam, and even then only because it's a mosque. More important to them is Al-Aqsa mosque which is no where near iconic in appearance, and that mosque is the one on the south side of the Temple Mount, built over Solomons Stables (named by the Crusaders who were the only ones to use the space as stables, and it has nothing to do with Solomon, it was built by Herod). Al-Aqsa is the one that they've tied to the story of Mohammad going to heaven. At least that's my understanding of it. The other thing is there really aren't that many structures on the Temple Mount, its mostly empty, which may or may not be a thing you are looking for. For Christians and Jews the entire Temple Mount is significant, because it's the Temple Mount, but there are like 2 or 3 churches (at least) in the Old City that are more significant for them because they have direct ties to Jesus and his life.
